【摘要】 目的:探討間歇性高糖誘導大鼠腎小管上皮細胞株NRK-52EA轉(zhuǎn)分化干預作用。方法:NRK-52E細胞分為空白組、正常葡萄糖組、高糖組、間歇性高糖組,分別作用72 h后,Western blot檢測轉(zhuǎn)化生長因子β1(TGF-β1)、Ⅰ型膠原、金屬蛋白酶-2(MMP-2)、α平滑肌肌動蛋白(α-SMA)以及甲狀旁腺素相關(guān)肽(PTHrP)的表達;CM2H2DCFDA試劑盒檢測活性氧(ROS)含量。結(jié)果:高糖組及間歇性高糖組NRK-52E細胞TGF-β1、Ⅰ型膠原、MMP2、α-SMA以及PTHrP表達均上調(diào);間歇性高糖組TGF-β1、MMP2以及α-SMA表達水平較高糖組高(P均<0.05),其ROS含量也顯著高于高糖組(P<0.01)。結(jié)論:間歇性高糖能通過氧化應(yīng)激誘導腎小管導管上皮細胞表達TGF-β1、MMP2,并促進NRK-52EA細胞轉(zhuǎn)分化為α-SMA細胞。

Effects of Fluctuating High Glucose on the Transdifferentiation of Tubule Endothelial Cell Line NRK-52EA in Mouse LIANG Hua-sheng,ZHONG Yu-huaDepartment of Endocrinology,The Ninth Affiliated Hospital,Guangxi Medical University,Beihai,Guangxi 536000,China)Abstract:Objective To investigate the effects of fluctuating high glucose on the transdifferentiation of tubule endothelial cell line NRK-52EA in mouse.Methods NRK-52EA cells were divided into control group,glucose group,high glucose group,fluctuating high glucose group,the expression of transforming growth factorβ1(TGF-β1),typeⅠcollagen,MMP-2,α-SMA and PTHrP were detected with Western blot,and the content of ROS was determined with CM2H2DCFDA kits after treated for 72 h.Results The expression of TGF-β1,typeⅠcollagen,MMP-2,α-SMA and PTHrP were all up-regulation in high glucose group and fluctutating high glucose group;the expression of TGF-β1,MMP-2,α-SMA and the content of ROS in fluctutating high glucose group was significantly higher than that of high glucose group(P<0.05 or P<0.01).Conclusion Fluctuating high glucose could induce the up-regulation of the expression of TGF-β1,MMP-2 in tubule endothelial cells through oxidative stress,and promote the transdifferentiation from NRK-52EAb to α-SMA.

糖尿病血糖間歇波動目前認為對于糖尿病并發(fā)癥的發(fā)生、發(fā)展起著重要的作用,其危害性甚至高于持續(xù)性血糖升高。腎小管上皮細胞轉(zhuǎn)分化是腎臟纖維化的重要病理基礎(chǔ),既往研究發(fā)現(xiàn)間歇性高糖能顯著促進內(nèi)皮細胞、心肌細胞纖維化進程[1-2],高濃度葡萄糖也可誘導腎臟纖維化[3],但間歇性高糖對腎小管導管上皮細胞的研究并不多見。本實驗將觀察不同條件葡萄糖狀態(tài)下大鼠腎小管上皮細胞中調(diào)控腎小管導管上皮細胞轉(zhuǎn)分化因子的表達,以探討糖環(huán)境對腎小管上皮細胞轉(zhuǎn)分化的影響醫(yī).學.全.在.線f1411.cn。
